I submit 30 - 40 directories each day and 3 days a week. I think huge submissions look unnatural.
I hope you know this quote "link building is not a rocket science". Go slow , it may take some time but surely you'll get your desired results.

My opinion is "Yes" no-follow links are improtant. Example: Think about digg or reddit these social bookmarking sites provide no-follow links but people share their stories though them just to get more traffic for website.
